アセットフォルダはルートパス(/)にある必要があります。 lib
フォルダーに保持する場合は、次のようにlib
をアセットの前に置きます。

lib/assets/fonts/Lato-Regular.ttf

補足:フォントを手動でインポートする必要がないように、 google_fonts パッケージをお勧めします。 Latoを含む何百ものフォントがあり、GoogleFonts.lato()
でアクセスして、_pubspec.yml
_ファイル経由でインポートすることなく、LatoフォントでデフォルトのTextStyle
を取得できます。
